No, because that's not how windows file replacement works.
Now when I press F2 in the overworld, it say 2.0.0.b4.2.1
Is it supposed to say that or is it supposed to say b4.3.0? Because is it patch 3.
(What I did is extract it from the download list and then put the extracted zip in the SMBX B4 folder)

You need to put the folder from the extracted zip named "data" into the Beta 4 folder. It will ask if you want to overwrite existing files, and you select yes.

Many of the items on the list are powerups, which cannot be created at this time.
Also I see many new playable characters on the list. We already have a large roster and that's even after they cut some characters. More characters means more balancing.
The tilesets from The Invasion can be extracted and placed in the custom 750 to 1000 range if you want to reuse them for other episodes. You can even use a tool in the editor to change them to png files to reduce bloat, like I did.
Actually, put THAT on the to-do list; convert all Invasion and Cliché levels to .lvlx and images to .png to reduce overall file count and size.

I'm not a developer so I don't get to decide what gets used and what doesn't, so there is always a chance something from that list gets put in. I was just pointing out that many items on the list were already in or exist in some form, while others were unlikely do to limitations or being unfeasible. It would also take some time to go through the list and link up each suggestion with its existing npc or physics pack where one does exist.
Earlier in this topic, there is a link to a roadmap spreadsheet showing what npcs the developers are working on for future builds.
If you want certain npcs, you could try to make them with LunaLua or get help in the LunaLua Help forum.

I've read Waluigi being split off from Ninja Bomberman has been considered in the past (as has splitting Yoshi from Klonoa) though this probably won't happen as we're getting Touhou, Celeste and Shovel Knight reps, and the other deleted character had their filter block ID replaced with a generic filter.

Waluigi and Yoshi would work better as their own but that’s just my opinion.
Shovel Knight and Celeste is cool but idk abou Touhou since that is more bullet hell like.

Literally most of the things you listened can be solved by simple graphic replacements like the Castle Goomba was hoch is just a Grey recolour. And some of these have been made into npc packs.

Literally most of the things you listened can be solved by simple graphic replacements like the Castle Goomba was hoch is just a Grey recolour. And some of these have been made into npc packs.

For the Red Star, do you mean the actual powerup, or the Power Star that makes said powerup appear in the Observatory? The former would require edits to the actual game code to fix a part that's basically FUBAR'd, but the latter can probably be done in LUA if custom variables to transfer flags between levels are possible. If they are, just make a graphics-swapped star that triggers an on-death event that does nothing, then in the level's Luna.lua have that event trigger a lua script to save some custom variable (I assume this is possible? Star coins save IIRC) that can be loaded to trigger an event on startup to enable/disable layers as needed. No idea what such code would be, though...

A rather long answer: show Considering that they were mostly suggesting Power-Ups, I'd say it's most likely the former.
Personally, I'd like to see more power-ups in this game, but considering how difficult it is to insert new power-ups ATM, and the fact that the team is focusing more on the fact that the next release will be a full release (instead of Beta 5), I have my doubts that we'd get new power-ups besides the ones planned to be in the full release.
Now, to bring up the latter, I'd like to see if this could be possible (not just with Red Power Star causing Red Star to appear, but also specific Stars causes specific Events (Boarding Bowser's Sub in Mario 64, for instance), and having different "keys" that opens different doors), because there's only so much you can do by limiting which areas Mario & Co. can enter with just locking it behind specific star amounts.
I'm not asking for an extension to Events that can make things like this possible; it'd be nice to have (especially for SM64-esque Episodes), but it's most likely not going to be implemented (correct me if I'm wrong).
TL;DR Version:
It's most likely the Power-Up, rather than the Red Star from SMG, and I'd like to see more advanced events, though I can understand it not being considered.
