To be fair, both Optimus Prime and Mega Man are probably immune to the neurotoxin and goo (assuming that it's poisonous) considering that they're robots.

Navi-"hey" "listen" "watch out" (would tattle(from LoZ MM) count too?)
Fi- haven't played SS but seen a LP of it to know Fi is definitely irritating
Starlow- nags where u should go next, sassy, and doesn't really like Luigi....

More maybe needed on that list...

Mega Man could teleport out of the facility - although it could be argued that GladOS could jam the signal of Mega Man's teleporting functions, rendering him unable to escape that way. I'm pretty sure Optimus Prime could simply crash through the ceiling and walk away, considering his size and strength. Regarding Mega Man against Optimus Prime, it can be said that while Mega Man has battled and defeated giant robots before, they were mostly controlled by simple artificial intelligences, while Optimus Prime is more sophisticated than those types of mindless machines. I'm not too sure about what Optimus Prime can do other than be really big and turn into a vehicle, but from the Transformers comics I read, he seems to carry guns that aren't part of him. He's still way bigger than Mega Man and even the type of giant mechas that he's faced before, so I'm not sure if Mega Man would defeat him with his standard buster gun. If we consider GladOS to have trapped them in a cramped facility, Mega Man has way smaller chances, depending on the weapons he has equipped.

I'd vote for Mega Man based on favoritism, but Optimus Prime is bigger and stronger. I'm not sure if GladOS can do anything other than cake jokes, though.

All three of them have already been done, Shrek and Hulk were last week and Yoda was a few weeks back. There might be repeats in future weeks (most likely specials or some other different Who Would Win) but as of now it's new every week. Thank you for the suggestion though!

Elsa and Anna are 2 characters from Frozen but how don't you know who are they. I don't know Lian neeson, though. Maybe a google search would help?
